Materials

Edwards Buffer

• Mature petunia plants with fully opened flowers
• 1.5mL Microcentrifuge Tubes (2x per sample; sterilized)
• Micropestles (1x per sample; sterilized)
• 1000uL Pipette Tips
• 200uL Pipette Tips
• 99% Isopropanol (ice cold)
• 70% Ethanol (ice cold)
• Sterile Distilled Water

Equipment

• Microcentrifuge (capable of 14,000 x g)
• Static Incubator (holding at 37C)
• Vortexer (set to max speed)
• Nanodrop or Alternative DNA Quantification

Step By Step

1. Weigh each 1.5mL microcentrifuge tube (one per sample) and write the milligram weight on the tube side.
