Aim

To attempt a pilot transgenesis run using carbide whiskers, petunia leaf disks, and pCambia2300-RUBYRED as the payload.

Results

No visible RUBY signal and there was an almost random distribution of dead explants, likely due to bleach-related necrosis. Must revisit properly.

Conclusions

After a long pause, I am rebooting this project and will first be focusing on some fundamentals prior to another attempt. I’ll focus on post-bleach survival of explants as well as the formation of callus since all of the protocols suggest embyrogenic callus being the ideal tissue for whisker tranformation. As a result I am closing this experiment and starting fresh.
