Step By Step

  1. Depending on your oligo supplier, either move the nanomoles decimal place written on the dry oligo tube over by one to the right (e.g. 19.4 nmols becomes 194) and add that many microliters of sterile distilled water to the dry oligo tube (19.4nmols oligo gets 194uL distilled water), or add however much your supplier tells you in order to achieve a 100uM final concentration. Elim Biopharma makes it such that I just need to add 100uL to each dry oligo tube to make the master stock solution of that primer.
  2. Vortex with 5 pulses of 1 second each, followed by 1 pulse of 5 seconds, and let the solution stand at room temp for 5 minutes.
  3. Spin down the tube briefly to collect any droplets to the bottom of the tube.
  4. Repeat step 2 and 3.
  5. Obtain and label one sterile 1.5mL tube for each primer you wish to dilute and add 180uL of sterile distilled water to each empty sterile 1.5mL tube.
  6. Add 20uL of the Primer Master Stock to the 180uL of distilled water in the corresponding tube.
  7. Vortex with 3 pulses of 1 sec and spin down briefly. This is your PCR Primer Working Stock tube, now diluted to 10uM, which is ready for use in PCR reactions.
