Ingredients

• Tris HCl 1M pH 8.0
• EDTA 0.5M pH 8.0
• RNAse A 20mg/mL (added to aliquots)

Equipment

• Magnetic Stirrer
• 15mL Serological Pipette
• 500mL Beaker
• 250mL Volumetric Flask
• 250mL Storage Bottle

Step By Step

1. To a 500mL beaker add 200mL of distilled water and begin stirring.
2. Add 5mL of EDTA 0.5M pH 8.0 to the solution.
3. Add 12.5mL of Tris HCl 1M pH 8.0 to the solution.
4. Transfer solution into a 250mL volumetric flask and top off the flask to the 250mL neck mark with distilled water. Invert several times to mix the solution.
5. Transfer solution to a 250mL bottle with a tight screw cap, label bottle, and store at room temp.
6. Aliquot 15mL of the Resuspension Buffer to a sterile 15mL tube.
7. Add 75uL of RNAse A 20mg/mL to the 15mL aliquoted resuspension buffer. Vortex to mix.
8. Label and date the aliquoted tube and store in the refrigerator at 4C. Use the aliquoted resuspension buffer for minipreps. DO NOT ADD RNAse TO MASTER RESUSPENSION STOCK SOLUTION. ONLY ADD RNAse TO ALIQUOT.
