Ingredients (needed quantity)
- Yeast Extract (5g)
- Tryptone (10g)
- Sodium Chloride (10g)
- Agar (15g)
- Distilled Water (1000mL)
Equipment
- Magnetic Stirrer and Stir Bar for 1000mL
- Analytical Balance accurate to 0.001g
- 1000mL Beaker
- Autoclave or InstantPot (Pressure Cooker)
- pH Meter (optional but ideal) or pH strips
- Autoclave Indicator Tape
Step By Step
- To a 1000mL beaker add 900mL of distilled water. Turn on stirring.
- Add 10 grams of tryptone powder.
- Add 5 grams of Yeast Extract powder.
- Add 10 grams of Sodium Chloride powder.
- Stir until fully dissolved. Adjust pH to 7.00 using 1M KOH if needed.
- Autoclave at 121C for at least 20 minutes. If using an InstantPot, set to pressure cook on HIGH temp and HIGH pressure for 30minutes.
